Jede neue wissenschaftliche Erkenntnis – sei es in Therapie, Diagnostik oder Patientenversorgung – bringt uns voran. Mal eröffnen sich neue Behandlungswege, mal gelingt ein besserer Umgang mit vorhandenen Ressourcen, und manchmal zeigt sich, dass ein eingeschlagener Weg nicht zum Ziel führt.
Die Therapielandschaft bei hämatologischen Entitäten verändert sich nicht nur hinsichtlich neuer Wirkstoffe, sondern auch durch Therapieoptimierung mit etablierten Strategien. Insbesondere für schwer behandelbare Patientengruppen wurden beim letzten Jahrestreffen der American Society of Hematology (ASH) neue Behandlungsmöglichkeiten aufgetan.
Grosse praxisverändernde Studien werden beim San Antonio Breast Cancer Symposium nicht erwartet. Nichtsdestotrotz wurden auch dieses Mal Studienergebnisse präsentiert, die zur Optimierung der Behandlung führen. Daten aus Phase-II- und Phase-III-Studien bestätigten Therapiestrategien insbesondere mit CDK4/6-Inhibitoren bei diversen Krankheitsausprägungen und weisen auf Konzepte zur frühen Behandlung von Brustkrebs hin.
At this year’s ESMO 2024 annual meeting, two pivotal phase 3 trials investigating pembrolizumab in advanced non-metastatic gynecologic cancers were presented, potentially changing the treatment landscape in both cervical and endometrial cancer.
Recent advances in breast cancer research have shown practice changing findings. This review article wants to highlight key insights from three major breast cancer trials presented at the last 2024 ESMO congress: the HypoG-01 trial, which evaluated hypofractionated radiotherapy in localized breast cancer requiring nodal irradiation, the KEYNOTE-522 trial, which investigated the addition of pembrolizumab to chemotherapy in localized node positive triple-negative breast cancer (TNBC), and the NATALEE trial, which assessed adjuvant ribociclib in hormone receptor-positive high risk early breast cancer. These studies contribute to ongoing developments in personalized treatment strategies across various breast cancer subtypes.
This study presents the findings of the first-in-human investigation of the mRNA-based cancer vaccine CVGBM in patients with newly diagnosed, surgically resected, and MGMT-unmethylated glioblastoma, (GBM), a group historically characterized by poor prognosis and limited treatment options (1).
Many interesting studies in the field of prostate cancer (PC) were presented at ESMO 2024. In my opinion three studies deserve to be highlighted: one in the localized setting, one in the metastatic hormone-sensitive setting (mHSPC) and one in the metastatic castration resistant setting (mCRPC).

Neoadjuvant therapies have the potential to increase R0 resectability and thereby improving surgical outcomes. Two recent phase II trials, PANDAS/PRODIGE 44 (1) and NICHE-2 (2), investigated neoadjuvant treatment approaches for borderline resectable pancreatic cancer and locally advanced mismatch-repair deficient (dMMR) colon cancer, respectively, with notable implications for clinical practice.
Since 2009, trastuzumab combined with chemotherapy has been the standard of care for HER2-positive metastatic gastric (mGC) and gastroesophageal junction cancer (GEJ), based on findings from the phase 3 ToGA trial (1). However, despite this initial success, the following decade brought a series of negative phase 3 trials, leaving first-line treatment options for HER2-positive gastric and gastroesophageal cancers without significant advancements (2–5).
At ESMO 2024 in Barcelona, various studies regarding the management of non-muscle-invasive bladder cancer (NMIBC), muscle-invasive bladder cancer (MIBC) and locally advanced / metastatic (la/m) bladder cancer were presented. This article aims to give a short overview of one outstanding study of each group.
In this article we review 2 studies on CD19 CAR-T cells, which were presented at the annual meeting of the European Hematology Association (EHA) 2024 in Madrid, Spain. The first study investigates the efficacy and safety of CD19 CAR-T cell therapies in ≥75 years old patients. The second trial explores if CD19 CAR-T cells expressing interleukin-10 (IL-10) are more resilient to exhaustion and ultimately show a better response rate in DLBCL or B-ALL.
Cancer of unknown primary (CUP) represents a heterogeneous group of metastatic epithelial or undifferentiated neoplasms where the primary tumour remains undetectable despite a standardized diagnostic evaluation. It is a rare condition accounting for less than 5% of all cancers and poses unique diagnostic and therapeutic challenges (1).

Seit September 2024 leitet Vincent Gruntz die Schweizerische Arbeitsgemeinschaft für Klinische Krebsforschung (SAKK). Nach einer Karriere in der Pharmaindustrie bringt er eine neue Perspektive in die Organisation, die seit 60 Jahren eine zentrale Rolle in der schweizerischen Krebsforschung spielt. Im Interview spricht er über seine Motivation, die grössten Herausforderungen in der klinischen Krebsforschung und seine Vision für die Zukunft der SAKK.
Die Identifizierung von prädiktiven und prognostischen Markern hat bei der Diagnose und Behandlung von gynäkologischen Karzinomen zunehmend an Bedeutung gewonnen. In dieser Übersichtsarbeit fassen wir aktuelle evidenzbasierte prognostische und diagnostische molekulare Marker bei gynäkologischen Karzinomen zusammen.
